Quick question. If someone is at the bottom of a ladder and a friendly charActer is at the top of the ladder can they mastermind to the person on the other end of the ladder. I am thinking no, but since the change of being able to attack up and down ladders, I am now not sure.

No. The only reason you can make close combat attacks up and down ladders is that the game permits it in the rules for them. The characters are not technically considered adjacent; thus, Mastermind doesn't work down a ladder, and neither can a character carry another character and set it down afterward on a different elevation at the bottom or top of a ladder or stairs.

No, you could not use Mastermind in that situation.

Characters on a ladder or stairs are only adjacent to the character on higher elevation for "close combat purposes only."

This is why you also can't use Poison with opposing characters in a similar situation, and why you can make a ranged combat attack against an opposing character in a similar situation.
