Can Silver Surfer carry two wildcards ( e.g. Spider-Man ) giving them the Defenders advantage ?.. and for the purposes of "Defenders, Ride With Me" would Silver Surfer still get the speed buff ( or would the carried characters need to have the actual Defenders team ability ) ?

Quoting the effects in question helps.

DEFENDERS, RIDE WITH ME: Silver Surfer may carry up to two single-base characters regardless of their combat symbols or possessing Battle Fury. When Silver Surfer carries only characters with the Defenders keyword or team ability, modify his speed value by +2.

"With" typically means they need to possess the team ability. Wildcards are out of luck.
In order for wildcards to benefit, it would have needed to say "characters using the Defenders team ability...".
