yes, everything would be blocked by barriers indoors

Outdoors it would depend on where the characters are located. Basically treat the giant character as at the same level as the other character. And then account for the Barrier normally. So:

A grounded barrier between a grounded giant character and a grounded standard character would block LOF.
A grounded barrier between a grounded giant character and an elevated standard character would not block LOF.
A grounded barrier between a grounded giant character and a soaring standard character would not block LOF.
An elevated barrier between an elevated giant character and an elevated standard character would block LOF.
An elevated barrier between an elevated giant character and a soaring standard character would not block LOF.
