1. Yes it does, you use it you lose it, just as any trap activated while set.
2. They will stay on the field
3. It doesn't matter how Makyura goes to the grave, it just has to get to the grave for hes effect to go off

for 1 and 2, you're basically still playing the trap card normally except it's straight from your hand and will do whatever its effect is.
For example
- Graceful Charity sends Makyura to the graveyard and you have a Seven Tools of the Bandit in your hand. You attack and your opponent responds with Mirror Force. Thanks to Makyura's effect for that turn, you can start a chain and activate Seven Tools of the Bandit straight from your hand, pay 1000 Lifepoints and negate the Mirror Force and let your attack succeed.
- Opponent's turn and they use Delinquent Duo. You end up discarding Makyura as one of your cards and you have a Trap Hole and Mirror Force in your hand. Your opponent summons a Don Zaloog. Thanks to Makyura's effect, you can activate Trap Hole from your hand in response and destroy Don Zaloog. The opponent then decides to attack you directly with the Luster Dragon #2 they have on the field already. You may then respond by activating Mirror Force straight from your hand since Makyura does not limit the number of traps you can play from your hand, but just the duration of how long the effect lasts, which is until the end of the turn it's sent to the graveyard.
For #3, it doesn't matter HOW it gets sent to the graveyard. Its effect is NOT optional, unlike Peten and Gilfer. The choosing of whether you wish to activate Traps from your hand is what is optional, so it can be sent to the graveyard in the middle of a chain and still be able to work for you. Just remember that it means you activate the traps STRAIGHT from your hand if you desire. It GOES NOT allow you set them and activate them in the same turn. If you set them, you have to wait a turn before activating them.
