Q) If I have Concrete Jungle in play, does my opponent have to be able to discard a card from hand to flip his location?
The scenario in which this appeared was that I had two Jungles face up and my opponent had only one card in hand. To flip his location, he would have had to discard 2 cards. I thought that this would mean he could not play his Doc Ocks’ Lab, but he disagreed.

Each of your opponents turns all locations he controls face down.
Ongoing: Whenever an opponent flips a location, that opponent discards a card from his hand.
It does not give your opponant an additional cost to play locations, it is a triggered effect that is put onto the chain. At the resolution of that effect, they have to do as much as possible. If they do not have any cards to discard, it does not affet their location in anyway.
Note again that it is not a cost, it is part of the effect. So, the reason they may flip the location is because they attempted to perform as much of the effect as possible, rather than trying to meet a cost payment and failing.

my question though, my opponent has the jungle, i flip lost city.. can i respond to jungles effect by using the only card i have on the lost cities effect for my sexy character thats about to win me the game?

As long as the character you're trying to pump with Lost City is involved in an attack, you can use the power in response to Concrete Jungle's effect, giving that character a +3/+3.
