Null Time Zone is played during the build phase and is only named upon resolution. That part I get. I suppose I don't quite understand the interaction between certain other cards.
Examples...
1. My opponent plays a plot twist that has a downside (discard a card, exhaust a character) and then passes priority, I then respond with NTZ naming that PT before it would resolve. What happens? Specifically, if my opponent plays Avengers Assemble!, discarding a leader card, then I play NTZ, does he lose his card, can he search? In general I am hoping for answers to this.
2. Now the situation is I play NTZ and my opponent decides to play something in hopes of getting it through...how many cards can you chain on your priority. What if my opponent decides to play every pump card in his hand before it resolves? What if I play NTZ, he plays MegaBlast, then I play another NTZ, then he plays Flying Kick. My intent would be to maximize my potential, and not be able for him to use his cards. If flying kick resolves, then my 2nd NTZ, naming MegaBlast, then what happens? Does MB still go off, and if so, why?
Firstly, Null Time Zone does not negate any plot twist that is already on the chain, it only prevents any plot twist with that name from being played from that point forward.
So, if you were to play NTZ in response to Avenger's Assemble, you can let NTZ resolve and name Avenger's Assemble, but he will still get the full effect (ie his search) from the original plot twist, however he could not play another copy of it this turn.
This should answer your second question to.
Remember, NTZ does not negate, only prevents a plot twist from being played. If it has already been played, NTZ will not stop it.

No, that would be a negation and NTZ does not negate it only prevents. But if he let NTZ resolve he could not play another Avenger's Assemble that turn. Some cards a player can play in anticipation to what you're going to call with NTZ, like Avenger's Assemble, Wild Ride, etc.

What can NTZ call out? Plot twists that are not ongoing only or anything that is not a ongoing PT like characters and Locations too. Wording was a little confusing.
