activate--target thunderbolts attacker or defender gets +2 attack while ready,
If I use this on a ready character then in the same attack exhaust my character does the +2 bonus go away since they are no longer ready or does it just check for readiness at activation.
The character is no longer ready, so yes, if it is during the same attack they lose the +2. One of the key drawbacks of the Thunderbolts is that so many of their tricks key off ready characters. But they do have Deadly Conspiracy to catch your opponent off guard. AND draw you a card. Everyone wins!! And by everyone... I mean you.
On a related note, if I play Team Tactics on my attacking Charcoal does he double his +5 or get no bonus at all? If no bonus, what if I use Deadly Conspiracy afterwords?
I'm thinking I need to play Team Tactics and then while it's still on the queue I play Deadly Conspiracy so that he's ready when the effect comes off the queue.

If you play Team Tactics on Charcoal in what context? If you are attacking with Charcoal you can't pay the cost of Team Tactics by exhausting Charcoal.
If you attack with Charcoal, once it is legal for you to use Team Tactics (ie when Charcoal is an attacker) Charcoal is exhausted. If you use Deadly Conspiracy to ready him after becoming an attacker and then exhaust him for the cost of Team Tactics then he will only receive +9 ATK as the card Team Tactics refers to the "exhausted" character's ATK as the bonus that the attacker receives. When Charcoal is exhausted his ATK is 9.
I forgot a Deadly Conspiracy in there, but Togorian is right about the scenario I was trying to set up. Sheesh! 4 Deadly Conspiracy, 4 Team Tactics, 4 Blind-Sided, 4 Charcoal, and 4 Songbird (6) = 20 cards of doom. Everything else in the deck can just revolve around getting these into play and maybe pumping them even more before the Team Tactics.
I tried this deck when MAV first came out. It rocks the house. Melissa is your win condition, and Marvels Most wanted combined with Win/Lose Deal, helps you get any card you need.
Another approach is to team up with the masters of evil, to use Black Knight and Ultron 5 for reinforcement breaking.
