And where do I know which location I can play multiple times and which not? Same for Ongoing plot twists.

you can play multiple copies of any plot twist which includes ongoing plot twists. In general Locations are Unique so you can only have one face up copy of any location in your resource row. Howere certain locations including Brother I Satelite and Checkmate Safe House say they are not unique and so you can have multiples flipped up. Starting in Marvel Team Up non unique locations like Mausoleum were given the non-unique version which also means you can have multiple flipped up.

All plot twists are not unique unless stated. The only Locations that are "non-unique" have it written in it's text box.

For example, Mountian Stronghold has the text:

Mountain Stronghold is not unique.
When Mountain Stronghold is flipped face up, search your deck for a League of Assassins character card, reveal it, and put it into your hand. Shuffle your deck and discard a League of Assassins character card.

Characters named Ra's al Ghul you control have reinforcement.
