Yep, you just have to 'break into' adjacency. Basically you have to roll a successful break away to move next to a giant. If it fails however, you're screwed.

But, that is only if you were already within the two square adjacency of the giant at the beginning of your action. If you are not, then a flyer can charge right in.

I hope we don't do this often, but this is going to be the second time tonight that Squirecam and I are gonna clash on the rules
A giant IS adjacent to a character with flight regardless of elevation or flight mode (Hovering or Soaring) and as such can engage in close combat from two squares away.
However, a character with flight ignores a character's base for movement purposes, (including giants). In other words, my Atom Smasher can charge your Wonder Woman from 2 squares. We agree on that. Wonder Woman is thus adjacent. As per normal rules for flyers, she must breakaway to charge. We also agree on that.
However, per normal rules for flyers, if Wonder Woman was not adjacent to any character at the start of her move, her ablitiy to ignore other character's bases for movement purposes supercedes any other rule, including the adjacency rules for giants, just as if she were to fly over a grounded non-flying character, a soaring flyer, and a giant, to charge my Atom Smasher at the end of her move.
I say this with the caveat that I could always be wrong, or perhaps I did not explain it correctly. Hopefully someone else will step in. It took me forever to type this post, so perhaps someone already has.

But,

From that, if a figure with flight begins its Charge move from more than 2 squares away, it may Charge into base contact.
If the Giant is already within two spaces at the start of the Charge, then the Charging character must break away to charge.
I take it that this has to do with Hulkbusters new power which states:
"Hulkbuster is adjacent to a giant character only when he is within 1 square of it."
This would lead me to believe that he cannot be based by Giants, but
can base them, as they are not adjacent to him, when he is two away.
This is cool for fighting Av Hulk who is a giant, and maybe the next
Hulk to be released in the M+M set will be a giant too.

When a grounded figure uses charge on a giant, they would normally need to stop 2 squares away and therefore can't make a close combat attack. Charge is useless this way. They would need to break AWAY to move closer to make the close combat attack.
Fliers don't need to stop when they become adjacent to enemies as long as theyre still moving; They can fly right through a square an opponent is in if they want to. A flier can ignore the "Giant's Grasp" and fly right in there and knock out some teeth.
As far as Hulkbuster. He isn't adjacent to giants at 2 spaces, but they're still adjacent to him. My figuring is that he ignores the 2 space limitation in that he wouldn't have to break away to close the gap. Since the power says nothing about giants being adjacent to him, Giants are still restricted to adjacency if Hulkbuster is within 2 spaces of them. They'd have to break, they can still make Close Combat Attacks on Hulkbuster.
