Also, what is the difference between buying booster packs and singles/gravity feed (other than the obvious fact there are more in a booster pack)? Does the variety or selection differ?

Yes, you will get different figures if you buy a booster of, say, Galactic Guardians than if you buy a gravity feed. Same looking figure, just a very different dial.
THe units section on this site is a GREAT way to look at these figs and see what you want to purchase.
Always remember to look at shipping costs before making a decision.

A gravity feed is a single booster set that usually has only 10 models in it. These are different from the ones that are in the actual set. Some sets only come in single pack boosters like the Avengers movie set came in single boosters then 3 piece team boosters.
As for the best place to buy them, I recommend locating a store in your area. The prices will be a bit more expensive but without venues to play at whats the point. If there are no stores in your area google search for the particular set you want and find the best price.
Also if you go to Heroclix.com you can look at the cards of the models from different sets. This will help you get an idea of what you want to start purchasing. The most recent sets are: DC: Superman, War of Light Fast forces, and The dark knight stuff. Marvel: Xmen fast forces, Galactic Guardians, Avengers Movie, and Incredible hulk. Those should be easy to find online for a reasonable price
and don't get caught up on blind (booster or GF) purchases. Go the singles route! IF you like Avengers then go to the units section, study the pieces and make informed purchasing decisions. There are lots of great pieces that are competitive and super-inexpensive.
You can pull a chase from a booster, but the smart play is to buy singles from a set that is out. OR save up and buy a brick or case. MM does free shipping on orders over $99.00, so you can get a brick of Cap and a brick of Superman for $100 and free shipping.
Thanks for all the help/info yall! I do have a local comic shop and a book store that KNOW about Heroclix ... the book store sells them, and the comic store has a place to play, but doesnt sell (as far as I know).
What is a "chase" ? Is that a rare or unique that is sought after?
I liked the selection on Troll & Toad ... but they have a ton of out of stock, and there shipping prices are ridiculous... I will check out the other one right away ...

Also a chase is an exremely rare piece. The rarity goes common which is like a grayish white color around the set emblem, uncommon which is green, rare which is silver, super rare which is gold and the card is light gold around the collectors number, and chases are gold and the card is gold around the collectors number.
Chases are technically "bronze", but just look like a dark gold. The exact rarity of chases varies from set to set, in most 60 figure sets it is usually 1 chase per 20 boosters. (9 times out of 10 though, price and playability aren't noticeably related, and 4 times out of 10 price and rarity aren't noticeably related on pieces)
T&T has progessively more reasonable shipping as you order more in 1 purchase.
Like everyone else, I advise you take the singles route to build your initial collection. Boosters are a nice purchase because of the excitement of cracking a blind booster and it introduces you to pieces you might not have given a second thought to. I recommend you play at least once at your local shop before buying a ton more. Find out if they are "themey" players who play very casual, comic book-/video game-accurate teams, or are they "cheesy" players who just run whatever pieces will get them the win. Then, decide which of these ways you want to play and look for the pieces that will help you build that type of team.

I would suggest going over the rulebook first. Then, I would spend a couple of hours perusing the meta-game thread to learn how different strategies work. From there, I would make an informed decision and purchase whatever I liked. This is what I did when I first started about a year ago. It's best to go for whatever figures you like best Many figures have some pretty awesome sculpts.
I will echo the other posters in that going the singles route at this point will be much better. You can pick and choose which pieces you want, based on personal preferences and themes (if any), without the hassle of getting a lot of pieces you don't want through a booster purchase. Sure, it is spendy if you want one of the super rare or chase figures, but that is the nature of a collectible game.
Now for future purchases, that is up to (again) personal preference and wallet capabilities. Nothing can match the fun of opening a brand new booster from a just released set and seeing a chase or super rare figure inside. Several in our group, due to both financial choices and the desire to cut back on mountains of duplicates they won't play, have gone solely the singles route, others buying no more than a brick and singles for what they don't get that way.
As people have posted Miniaturemarket and ############.com are good and tend to have the most product, both sealed and loose.
Here are some of my favorites too:
A place that most people do not think about is www.scifigenre.com. They have really good prices on FF and Sealed product. They do not sell singles. They have free shipping for $75.00.
For loose figures try www.strikezoneonline.com they do not sell sealed only singles. Free shipping on orders over $40.00, orders below that ship for a dollar.
I am not sure about buying singles just because I am new enough that I am not certain what I want or could compliment my strategy yet. Hmmm ... I think it is totally cool that there are places that sell singles though so that you CAN build your you collection as you like. I think I need to play some more before I build some serious strategy and buy accordingly.
I think one of the things that attracted me to the HeroClix experience though are the boosters ... its cool not knowing, and there is always that chance for a chase or another nice figure.
Its a bummer that I live in Vermont where there is virtually NO PLACE to get this stuff around me. I know of one small local place 15mins away with MINIMAL selection, and then the next closest I know about is an hour away ... with gas prices the way they are, I might as well go online. And finding people who know about the game or even play is even HARDER! I recently showed a friend of mine the game and he and his son are interested in playing with my son and I ... but THATS IT! lol

I was just wondering, have you tried the Wizkids event system to find a location near you? It's pretty useful for finding venues/tournaments near you If there aren't any locations that sell Heroclix near you, that sucks
