There are special rules for the Kill and Keep that modify the normal Dropship rules. Go to the Wizkids site and cllick on the Kill and Keep button to learn more on those rules.
In order for the game to end, I imagine that all the players would have to be wiped out, the BM's forces PLUS dropship would have to be destroyed, or there is a suggested time limit of four to six hours.
In order to "make your move" on the mechs you want, you have to wait for the unit you want to kill to be deployed by the BM if it isn't deployed at the start of the game. Then, you'd want to damage it fast and hard ASAP in order to get the kill credit for it. Standard tactics of waiting for things to get into your Assault Order range and then blowing them up apply for this event.
If the Dropship is destroyed, the BM cannot deploy more units (since there are no more Mech bays and ramps to deploy from. If that happens, the only units the BM can use are the ones they still have on the field. If they are destroyed, then the game has to be over, since one side no longer has an able unit.
