1) Yes... to an extent. It'll explain everything that would come up in most games. The exceptions are special rulings on card and in depth rulings such as how attacks are entirely declared.
2) Yes. DC Set comes out middle of July, and will feature Batman VS. Joker as a Starter- with Teen Titans as a team that can be found in booster packs.

Marvel Origins has both a starter, featuring the X-Men vs. the Brotherhood, and boosters out, containing more of the aforementioned teams and cards for Dr. Doom, Sentinels, the Fantastic Four, and others.
Spider-Man has a starter out, featuring Spider-Friends vs, the Sinister Syndicate. They don't have boosters out yet, but they wanted to get the starter out before the movie hits.
DC Origins is coming out soon, and will have a starter featuring the Gotham Knights (Batman and co.) vs. the Arkham Inmates (Joker, Riddler, etc.). Boosters will contain additional cards for these teams as well as the Titans, the Fearsome Five, and the League of Assassins.
The basic rulebook in the starter will, as Latuki Joe mentioned, cover the basics, so if you buy a starter you will be able to get playing right away.
If you go to the Upper Deck site, you will be able to find a comprehensive rulebook as well as errata for some cards, along with an online demo that may help you understand the rules a little better.
The game is not Marvel or DC. It is fully compatible and tournament play supports any and all cards, so you may look at getting some Marvel cards even if you only want to play DC. You will be able to trade off your unwanted cards to help you get more of what you want, and this site has a trading post to help you do just that if you don't have enough players in your area.
