I just started DC Vs. Picked up a starter and a few boosters. I wanted to make a Gotham Knights and League of Assassins deck. But also curious over all to see what Teams/Cards are must have for DC. And also, I have a few "My Beloved"/"World's Finest" cards and was thinking of a Knights/Assassin deck. What cards (standard 60+ card deck) should I make? Thanks for your help in advance.

If you're looking to play on a budget (which is at least as much fun as getting all crazy serious about it), the League of Assassins pretty good to play-- most of their really cool stuff is Uncommon, and their best characters are Common.
With the Gotham Knights, their /best/ card is Fizzle, but it's one of those money-rares that can be a bit difficult to collect. However, they have several consistently good characters that can work together very nicely.
It sounds like, based on your collection, that one deck that will let you opportunistically collect both teams would be the way to go.
You definitely want a playset (4 copies) of:
Batman, Caped Crusader [recruit 5]
Ra's Al Ghul, Master Swordsman [6]
Tim Drake, Boy Wonder [2]
Kyle Abbott, Wolf in Man's Clothing [3]
Huntress, Helena Rosa Bertinelli [3]
Bane, Ubu [5]
Catwoman, Selina Kyle [4]
Lazarus Pit
Mega-Blast
Combat Reflexes
Clocktower
My Beloved
These are commons-- most anyone playing in your area will cheerfully trade you enough to fill out your set for something nice that you're not using. The main thing is to stay away from characters that have Loyalty, as they can be very hard to play in a team-up deck.
You want to get as close to a playset of:
Mountain Stronghold
Bat-Signal
Utility Belt
These cards really good, but they are Uncommon, and a little harder to get your hands on.
Round out your collection with:
Flying Fortress (2 copies)
Lady Shiva, Sandra Wu-San [5]
Dr. Tzin-Tzin, Master of Hypnosis [4]
Ra's Al Ghul, Immortal Villain [4]
Malaq, Money Man [1]
Dick Grayson, Defender of Bludhaven [6]
Spoiler, Stephanie Brown [1]
Azrael [6] (but you usually play him at 7)
Sadly, Dr. Tzin-Tzin is more useful than Ra's [4], because of that pesky Loyalty requirement.
Then you can chase your rares:
Lady Shiva, Master Assassin [7]
Ra's Al Ghul, the Demon's Head [8]
Superman, Big Blue Boy Scout [8]
Alfred Pennyworth, Faithful Friend [1] (2 copies if you can get them)
There's a mix in there of those cards that should make for an interesting deck. Beyond that, just trade opportunistically for just about anything that is Batman- or League of Assassins-affiliated (except maybe Clench Virus and Batman), and mix them up into your collection.
The theorists say you're supposed to stay as close to 60 as possible, but playing for fun-- you may end up with more. That's okay in my book. <shrug>
Glad to see your coming to the CCG world. As for your deck concept, you'll want to have a min. of 3 combo cards in your deck (to allow all of your characters to work together for Team Attack/Defense if need be) then you'll want to build a concept for the deck, do you want it to be fast drawing, poision or just quick beat down.

ericlui gives a lot of good ideas. I however, like to mix my Knights up a bit more.
3 drop Batman if you can nab it, is sweet. Him and 4 drop Ghul can shut down someone on the early turns. I also like Josef over Malaq for League's 1 drop, dunno which is rarer since my box had a Josef and 0 Malaq. Josef is especially nice with discard effects (Greater of Two Evils, Catwoman, etc). Also, if you don't want to go with big name guys like Batman and his pals, try the GCPD swarm with Commish Gordon, Bullock, and the HQ. Could get some interesting plays in an army deck. And the PD has some nice stall ability. Other League Army after the team up would also get Gordon and Bullock's powers.
