Can you activate this card if you have a face-up Karakuri monster in attack mode and is changed to defense when it is attacked by your opponent's monster? The ruling was not really clear to me. thanks.

no because the attack target was selected when the karakuri was in attack mode not when it was in defense mode.

Just based on the wording, the Karakuri monster must already be in face-up Defense Position right at the time it is selected as the attack target.

Unfortunately, the only ruling listed is that it can work when you attack your opponent's face-up Defense Position Karakuri monster.
